Description:
The AQ-08 photometer contains the optical engine and other hardware elements required to measure absorbance at 260 and 280nm. Unlike other devices used for the quantification of DNA, RNA, and protein, the AQ-08 uses a 260nm LED for measuring ssDNA, dsDNA, and RNA and a 280nm LED for the quantisation of protein. Both LEDS are used to generate the 260/280nm purity ratio. The value of using LEDs for these methods include; long lifetime without the need for replacement, low heat production shielding samples from degradation, and high photometric precision. In addition to the LEDs, optical engine of the AQ-08 has the 260nm and 280nm filters along with photodiode detectors. This optical engine yields a reading range from 0.0 to 4.3 OD allowing measurements of less than 1ug/mL ds DNA. When a measurement is initiated the optical engine passes over the 8 sample vessel and measures at each position. The results are sent to the graphical user interface in an easy to understand format.
How it works?
There are three main elements of the AQ-08, the AQ-08 photometer , the 8-channel sample vessel, and the graphical user interface.
The 8 sample vessel
The vessel is designed to hold eight separate samples each in its own individual well. Since each sample is housed separately, recovery of your sample is simple and insured to be free of contaminants. This simply design allows for you to add or remove samples with standard or 8 channel pipettes.
The Graphical User Interface:
The AQ-08 Eight Channel nucleic meter is a PC operated device with an intuitive user interface. Simply launch the user interface and you are ready to go. This powerful yet easy to use interface allows you to enter sample name, enter a dilution factor, or select an assay type. After blanking, simply click Measure sample and the AQ-08 does the rest. The results sent to the user interface will show raw 260nm and 280 nm OD, the sample purity, and the concentration. The concentration is calculated based upon the respective OD, the extinction coefficient for the samples type and the dilution factor entered. Upon completion, click the export export function and the data is saved in a .CSV file format which is easily opened with Microsoft Excel ™.
